
Our course on Downstream Processing teaches the essential skills needed for protein purification and analysis. In this course we will cover the preparation of protein buffers and explore cell harvest and lysis techniques, understanding the importance of temperature control and different centrifugation methods. As we dive into chromatography, you’ll learn about various methods such as affinity capture and size exclusion, along with sample analysis techniques. You will gain hands-on experience in sample preparation, equipment maintenance, purification strategies and troubleshooting methods.
Course Topics
- Overview of the protein purification process, a typical workflow, and related terminology.
- Harvesting methods for bacteria vs. mammalian cells, gaining practical experience in centrifugation, chemical lysis, and microfluidizer processing.
- Chromatography methods and columns: affinity capture, ionic exchange, size exclusion.
- Components and methods for the preparation of protein buffers.
- Sample preparation for each step of the purification workflow.
- The AKTA Pure 25 Chromatography System:
- System set-up
- Executing methods
- Basic maintenance
- Handling and maintenance of chromatography columns
- Methods for buffer exchange and protein concentration.
- Sample analysis: SDS PAGE, and protein quantification using the Bradford assay and absorbance measurements.

- Audience: This course is intended for participants in biomanufacturing and biotechnology with no previous biology or biomanufacturing experience. Participants are required to be 18 years old or older, and have a high school diploma or equivalent experience.
